Most oncology targets come from cancer cell lines and CRISPR screens, and many of them do not hold up in patients. A tumor's genome shows what the tumor could afford to lose. Genes it depends on are rarely lost; genes it does not need are lost freely. We measure these losses down to a single gene copy and score every gene in every tumor type. We map which genes cancers depend on, using patient tumor genomes instead of cell lines. The maps are built from roughly 14,000 patient tumors across 26 cancer types and cover ~20,000 genes. Partners use the maps to: - validate a target before committing to a program - choose the tumor types where a target matters most - find co-dependencies that limit monotherapy response - estimate a program's probability of success before the next trial - pick target pairs for bispecific antibodies Targets found with the maps are already licensed to industry partners. We are also building our own pipeline of bi- and tri-specific antibodies for pancreatic (PDAC), colorectal and non-small cell lung cancer.
