Riche d'une longue histoire, l'Université Panthéon-Assas est l'héritière de l'ancienne Faculté de droit et de sciences économiques de Paris, composante de la Sorbonne, installée au cœur du quartier latin depuis le XIIIème siècle. A cette époque, l'Université de Paris comprenait quatre Facultés : théologie, médecine, arts et droit. Pendant près de cinq siècles, l'enseignement juridique a porté exclusivement sur le droit ecclésiastique, jusqu'à ce que Colbert introduise, sous le règne de Louis XIV, l'enseignement du droit civil pour donner ainsi naissance au droit français.
Le siège principal de l'Université est installé place du Panthéon, dans le 5ème arrondissement de Paris. Construit en 1770, le bâtiment destiné à la Faculté de droit s'inscrit dans le projet d'urbanisme des abords du Panthéon, alors église Sainte-Geneviève, confié à Soufflot. Devenu mairie sous la Révolution, il retrouve son affectation d'origine dès 1805.
Le Centre de la rue d'Assas est construit au début des années 1960 par J. Le Maresquier. Il donne son nom à l'Université Panthéon-Assas, créée après les événements de mai 1968 ayant conduit à l'éclatement de l'Université de Paris.
