Glucagon-like peptide-1 (GLP-1) receptor agonists have revolutionized the treatment of type 2 diabetes (T2D) and obesity, but they require lifelong administration and face high discontinuation rates. Treatment withdrawal triggers disease relapse and progressive weight regain. Paltech has developed an innovative solution to restore the defective glucose sensing pathway from the periphery to the brain. At the core of the innovation lies a breakthrough understanding of peripheral vagal glucose sensing mechanisms. Paltech restores expression of the GLP-1 receptor (GLP-1R) on vagal nerve afferents along the portal vein—a critical mechanism impaired in T2D and obesity. Utilizing a long-acting implant, Paltech therapy lasts several years after a single, swift laparoscopic procedure. Since the drug is not released systemically, it eliminates unwanted effects. The minimally invasive AI-guided laparoscopic procedure integrates into existing clinical workflows. Preclinical studies in a large diabetic animal model demonstrate glycemia normalization, restoration of dynamic insulin secretion, insulin resistance reversal, and sustained weight loss without sarcopenia. Paltech is preparing for FDA interactions and first-in-human studies, engaging partners and investors to redefine T2D and obesity treatment toward long-term remission—without cumbersome administration.
